the royal key also called the ouken is the key to open to the Kings realm, Aizen wants to kill the king, so he can stand alone in soul society.
I personally think Aizen is gonna get the Ouken by getting it out of Yama, who knows the the location of the true ouken, I don't think he has the means to destroy karakura town.

Yamamoto said Aizen's goal is to create the ouken, enter the royal dimension, and overthrow the King of Soul Society. I believe he means that the King of Soul Society lacks the power to be considered "standing atop the heavens", whereas Aizen lacks the authority to be given that title. Therefore, Aizen will strip the King of his authority by force, and thus fill the "vacancy on the throne in the sky".

Aizen has Gin, Tousen and the three remaining Espada (if Ulquiorra didn't manage to make it). If he can't defeat the Gotei 13 and the rest of Earth's forces to gain his 100,000 souls, then I believe Aizen would have to resort to getting the key from Yamamoto as well. But even that is looking less likely...

I thought that too, I believe it was mentioned the King was missing? Regardless Aizen still needs the key to get into his realm.
